ObjectiveTo detect the expression and significance of Transcription factor Snail and Matrix metalloproteinase-7 in lumbar intervertebral disc.To discuss the role of Transcription factor Snail and Matrix metalloproteinase-7 in the course of the intervertebral disc degeneration and clarified the mechanism of the lumbar intervertebral disc degeneration and prolapse.MethodsForty three lumbar IVD paraffine specimens were collected from Department of Orthopedics ,the First Affiliated Hospital of China Medical University during the period from February 2006 to Novermber 2006.Experiment group contains thirty two prolapse intervertebral disc and control group contains eleven discs which collected from lunbar spine fracture and scoliosis. Experiment group was divided into protrusion and extrusion type and sequestration type.Specimens were fixed in 10% formalin and were embedded in paraffine.5um serial section were cut and SABC immunohistochemistry method was used. Transcription factor Snail and Matrix metalloproteinase-7 antibodies stain the section and Gray Scale Value were collected by using microimage analysis system.Using software SPSS12 for statistical analysis.Means SD for results of analysis of specimens were calculated and compared using Student t test. ResultsTranscription factor Snail and Matrix metalloproteinase-7 is expressed in cytoplast and cytoplasm.The expression of Transcription factor Snail in the experiment group is lower than in the control group,and the average gray value of microimage analysis system and immunohistochemistry between them has statistical significance,but no statistical significance between protrusion and extrusion type and sequestration type. The expression of Matrix metalloproteinase-7 in experiment group is higher than in the control group,and the average gray value of microimage analysis system and immunohistochemistry between protrusion and extrusion type and sequestration type has statistical significance.ConclusionsThere is obvious correlation between the expression of Transcription factor Snail and Matrix metalloproteinase-7 in lumbar intervertebral disc and lumbar intervertebral disc degeneration and prolapse.The decrease of the expression of Transcription factor Snail and the increase of the expression of Matrix metalloproteinase-7 may be the cause of the lumbar intervertebral disc degeneration and prolapse.
