Objective:1 To establish the multifactorial model of necrotizing enterocolitis of neonatal rat using artifical feeding, hypoxia and cold stimulus and evaluate on it.2 To study the change of platelet-activating factor and tumor necrosis factor-a density in neonatal rat model of necrotizing enterocolitis,and analyses the correlation between the PAF> TNF-a level and the scoring of intestinal injury.Methods:According to 2×2 factorial experiment design,32 neonatal Sprague-Dawley rats.provided by institute of medical sciences of Fujian Province, were randomly divided into 4 groups (A,B, C, D)with 8 rats in each group and without differentiation of gender.Group A,artifical feeding, hypoxia cold stimulus;Group C,hypoxia cold stimulus Group B,artifical feeding,Group D,normal contral.NEC model was established as following method:24 two-day old SD rats wete part from their mothers and 100% N2 was administered into an airtight container,then 24 rats with NEC were put into and stayed for 90 seconds in the container when the concentration of oxygen in the container was read 0 by oxygen detector,and then the rats were put into 4℃ refrigerator cold stimulus for 10 minutes.Repeat this procedure twice daily with an interval of 12 hours for 3 days,On the 4th day.all rats were were killed to cut out 2cm of ileum ends and cecum, fixed in 10% paraformadehyde, embedded in paraffin, microtome-sectioned at 5 um slices, and...
