PARTⅠAnimal model establish and protective effects of curcumin on neonatal rats with necrotizing enterocolitisObjective: On the basis of animal models of NEC domestically or abrord, establish NEC model and objectively evaluate protective effects of curcumin on neonatal rats with NEC.Methods: Separated from maternal after 24 hours of birth, SD rats were fed with rat milk substitute and undergone 100% N2 hypoxia 3 minutes and 100% O2 hyperoxia 3 minutes,then were given 4℃cold exposure 10 minutes. Forty neonatal rats were randomly divided into 4 groups(n=10 each): normal control, solvent control,NEC model,curcumin intervention. The general situations and weights of the rats were observed for 3 consecutive days, and the rats were sacrificed on the 4th day. Intestinal tissues were obtained for examining the gross morphological changes,morbidity,histopathological changes and scores.Results: Compared with normal controlled group, the general situations of NEC model group were poor and findings in intestinal gas,hemorrhage and necrosis are obvious. The Body weight changes and pathology scores of NEC model group were higher than normal control group (P <0.05). Curcumin treatment ameliorated the general situations and histopathological signs in rats with NEC, reducing its incidence. The body weight changes and pathology scores were significantly different from the NEC model rats(P <0.05).Conclusions: Artificial feeding plus hypoxia hyperoxia cold stimulation is an ideal modeling method of NEC. The intestinal tissue damages are consistent with the clinical manifestations and pathological changes of human NEC. Curcumin has protective effects against NEC in neonatal rats.PARTⅡPrevention mechanism of curcumin on neonatal rats with necrotizing enterocolitisObjective : This Study examined the effects of curcumin on necrotizing histopathological changes, COX-2 and HIF-1αexpressions, TNF-αand IL-10 concentrations in neonatal rats with NEC,in order to investigate the prevention mechanisms of curcumin against NEC.Methods : Forty neonatal rats were randomly divided into 4 groups(n=10 each): normal control, solvent control,NEC model,curcumin intervention.The general situations of rats were observed for 3 consecutive days, and the rats were then sacrificed on the 4th day. Intestinal tissues were obtained for examining the histopathological changes,COX-2 and HIF-1αexpressions,and TNF-αand IL-10 concentrations.Results : Curcumin treatment ameliorated the general situations and histopathological signs in rats with NEC. TNF-αand IL-10 concentrations in the NEC model and the curcumin intervention groups increased significantly compared with those in the normal and solvent control groups(P <0.05); The concentration of TNF-αdecreased (P <0.05),while the concentrations of IL-10 increased significantly in the curcumin intervention group compared with the NEC model group(P <0.05). Immunohistochemistry results indicated that positive expressions 0f COX-2 and HIF-1αin the curcumin intervention group were significantly higher than that in the normal control group and the solvent control group (P <0.05), while lower than that in the NEC model group (P <0.05).Conclusions: Curcumin has protective effects against NEC in neonatal rats, possibly through inhibiting COX-2 and HIF-1αexpressions , reducing TNF-αcontent, ane increasing IL-10 content.
