Objective:In order to investigate the prevention and treatment of Grifola frondosa and its immunological mechanism against type 1 diabetic mice, we studyed the changes of type 1 diabetic mice Th1/Th2 type cytokines ,blood sugar of serum and mice behaviours. Methods:40 kunming strain mice were randomly divided into 4 groups: normal control , model group , model treated with high dosage of Grifola frondosa , model treated with low dosage of Grifola frondosa. The diabetic models were established by abdomen injection with alloxan .Whether the model were successful ,we confirmed it by observing the changes in mice behaviours, levels of blood sugar and IL-4, IL-5, IFN-γ in serum. Model group were administered 0.4ml NS per mice everyday; model treated with different dosage of Grifola frondosa were daily administered 0.4ml drug solution of 100%and 50% concentration respectively. One mounth later,the levels of IL-4, IL-5, IFN-γ were measured by ELISA. The glycemia were determinated by GOD method.Results: Diabetic models were developed successfully, when the symptoms of polydipsia,polyuria,polyphagia, weight loss were present. As for model treated with Grifola frondosa,the symptoms were mitigated,while control group were normal . The levels of blood sugar and IFN-γ were increased very much obviously in model group (P<0.001, P < 0.05),compaired with that in control group. The levels of glycemia and IFN-γ were reduced very much significantly in high dosage group of Grifola frondosa (P<0.001, P<0.01), compaired with that in model group.The levels of IL-4, IL-5 in serum with high dosage of Grifola frondosa were raised in comparision with that in model group(P<0.01, P<0.05).However,these was no marked difference in levels of IL-4, IL-5 between model group and low dosage group of Grifola frondosa (P>0.05).Conclusion:Grifola frondosa had a beneficial effect on reducing levels of blood sugar in type 1 diabetic mice.It can also lower significantly levels of IFN-γ and enhance levels of IL-4, IL-5 in serum.In brief,The ratio of Th1 and Th2 were influnced by Grifola rondosa ,which maybe play an important role in prevention and treatment for type 1 diabetes mellitus(Type 1 DM).
