Background and Objective The goals of this study were to label the antisense oligodeoxynucleotides(ASODN) indirectly by Chloramine T method and Iodogen method, then to discuss the labeling yields,radiochemical purity,specific activity and stability in different conditions. We discussed the right labeling method and conditions in order to lay the experiment foundation of the antisense medicament labeled by iodine for clinical application.c-erbB2 active and amplification were found in earlier period of mammary neoplasm. Oncogene active result in cell mRNA overexpression. ASODN can identify mRNA and specially conjugated it. Radiolabeled ASODN was designed complementation to target mRNA for imaging changes in the level of gene expression in vivo. It can realize the earlier period and no-wound diagnosis for neoplasm.It is obvious that labeling the ASODN by radioelement is the most pivotal process. Imaging experiment in vivo and vitro can't not be carried out if the labeled medicament preparation didn't succeed. Therefore, it's significant for the clinical application of antisense technology that searching the availability,convenient and stable labeling method.Methods1. Label the Ethylenedicysteine(EC) by Chloramine T method. Testify the feasibility of 125I labeling EC.2. Design and compose the part sul-modify ASODN of c-erbB-2 gene. ASODN sequence was 5'-CTC CAT GGT GCT CAC-3'.
