PARTⅠPREPARATION AND QUALITY ASSESSMENT OF PACLITAXEL-CARRYING MICROBUBBLEObjective: Self-made liposome microbubbles containing paclitaxel (PLM) were developed and evaluated as a new ultrasound contrast agent for chemotherapeutic drug delivery.Methods: PLM were prepared by a shaking device for 30 s. On the 1st and 12th day, characterization and physical properties (concentration,shape,size,surface potential,pH value) of this microbubble were detected by light microscope and Zetasizer 3000HS,respectively. The entrapment efficiency (EE) (1st and 12th day) and drug–loading amounts of paclitaxel was determined by reversed phase high-performance liquid chromatography(RP-HPLC). Lipospheres containing sudan black dye were produced to demonstrate the acoustically active liposphere-ultrasound release concept. Drug release of PLM destructed by ultrasound were observed in vitro spectrophotometrically. Acute toxicity testing was performed in mice comparing Taxol to PLM. The acute toxicity reaction of...
