In 1997, Japanese scholar Kuro-o discovered the KLOTHO gene in the study of genetic variation of hypertension, the absence of which may contribute to a mouse's aging. Ever since then, scholars from different nations have made researches on this gene. Meanwhile, with the completion of the human genome project (HGP), scientists paid increasing attention to the SNP, the genetic marker to locate pathological gene. The frequency of SNP and the association mode between SNP (haplotype) are the key to explain a series of unbalance between the SNP and candidate gene. Objective: (1) To establish PCR reaction system that uses allele-specific primer PCR technique to detect SNP of KLOTHO gene. (2)To analyze the genetype distribution of three single nucleotide polymorphisms (SNP) sites of KLOTHO gene in different age groups and the relation to life span. (3) To study the association between the three single nucleotide polymorphisms (SNP) sites and several kinds of diseases. Methods: To extract the genomic DNA of leukocytes from 640 individuals, who are divided into two groups, the aged group and the youth group. The mid-old age samples are from the physical examination department in the first affiliated hospital of Chongqing University of Medical Sciences, and the youth samples are from the physical examination of freshmen in a university.
