There are 56 nationalities in China,among them 26 are distributed in Yunnan Province and 15 nationalities including Nu and Dulong ethnic groups inhabit in Yunnan only.Owing to geographic,traffic and historical reasons, these two nationalities have been keeping in a relatively isolated state and therefore isolated groups were formed.A collection of genetic samples and investigation of medical and educational status were conducted among Nu and Dulong ethnic groups in all villages in Bingzhongluo and Dulong-River regions in 2003,November 2006 and April 2007.Through our investigation,we knew the distribution of the main disease in the two ethnic minority regions,the eradication of malaria and tuberculosis are still the arduous tasks of the future,and the local AIDS also began to spread.Yunnan province is China's main malaria-endemic areas,its incidence of malaria in the nation is No.1,and its total number of cases more than 30 percent in the nation.Yunnan malaria mainly concentrated in the western and southern border regions,generations living in the Nu-River in Yunnan are in the high incidence area of malaria.In recent years,a number of studies found both at home and abroad,NOS2 gene mutation may play a role in resistance of malaria.NOS is a key enzyme of NO catalytic synthesis.At present,people found that human NOS gene can be divided into three types:Typeâ… is also known as the brain-NOS(bNOS),the nerve-NOS(nNOS)or NOS1.Typeâ…¡also known as inducible NOS(iNOS)or NOS2.Typeâ…¢,also known as endothelial NOS(eNOS)or NOS3.NOS2 gene can produce large quantities NO by cytokines,endotoxin inducement,and involved in the pathophysiology of a variety of disease processes.Nitric oxide synthase(NOS)catalyzes the reaction of L-arginine into NO.NO,as a liberal diffused cell messenger molecule,in the neurotransmitter transmission,vasodilation regulation,infectious diseases and tumors protective immunity plays an important role.Many experiments proved that NO in the protection immune of Plasmodium falciparum infection plays an important role.NOS2 gene promoter region of the G-954C,C-1173T two SNP loci with the occurrence of malaria have some relevance.We selected 84 cases of healthy people as studing object in Nu-River,Yunnan.We extract genome DNA,use nested allele-specific primers PCR technology to detect the NOS2G-954C SNP and the NOS2C-1173T SNP, and then use agarose gel electrophoresis technology to detect the products of PCR reaction,and use the gel imaging device in the UV lamp to observe and take picture.The results showed that:In the 84 cases studied,NOS2G-954C SNP genotype frequencies were:G/G 63.10%,G/C 32.14%,C/C 4.76%,allele frequencies were:G 79.17%,C 20.83%;NOS2C-1173T SNP genotype frequencies were as follows:C/C 58.33%,C/T 30.95%,T/T 10.71%,allele frequencies were as follows:C 73.81%,26.19%T.
