Objective:To explore the effect on bile transport in the placenta of intrahepatic cholestasis of pregnancy(ICP)by detecting the expression of farnesoid X receptor(FXR) and MDR3mRNA in placenta and examination the effects of Chenodeoxycholic acid(CDCA)or Ethinylestradiol on the expression of MDR3 and FXRmRNA in placenta.Methods:25 case of ICP and 25 case of normal pregnancy as compared group by randomized choosed.The expression of FXR and MDR3mRNA in placenta were detected in both groups by RT-PCR. randomized choosed placenta of 37-40 weeks gravida by cesarean section. we first treated human trophoblast with diferrent EE (0,2,4,6,8,10,12uM), CDCA(0,20,40,60,80,100uM),UDCA(100,200,300,400,500,600,700uM) for 24 hours and with 80 uM CDCA for 3,6,9,12,24,30 hours,then analyzed endogenous FXR,MDR3mRNA expression by RT-PCR. We treated human trophoblast of intrahepatic cholestasis of pregnancy with diferrent EE UDCA(100,200,300,400,500,600,700uM) for 24 hours,then analyzed endogenous FXR,MDR3mRNA expression by RT-PCR.
