Objective: The intramembranous pathway of amniotic fluid absorption has been recognized as a critical regulatory path for amniotic fluid resorption, which contributes importantly to amniotic fluid volume homeostasis, but the underlying molecular and cellular mechanisms for water absorbed across the amniotic membranes remain unknown. The aquaporins are membrane-spanning proteins in many cells types involved in water transport. The current study sought to investigate expression, location and regulation of aquaporin 8 in human amnio-derive WISH cells and to explore the mechanisms for water absorption across the amniotic membranes and study the feasibility of regulating AQP8.It is hoped to find a new strategy to guide the treatment for disorder of amniotic fluid volume in clinic.Methods: (1) Human amnion-derive WISH cells were cultured. The cells were divided into control groups and study groups at random, which were examined at interval time of 2,4,8,16,24,and 48h, respectively (n=6). The cells of the control group were cultured in normal culture medium. The study groups were established by exposed to various osmotic mediums.
