Background Viral vector and nonviral vector are two main category of shuttle vector for central nervous system gene therapy. They still couldn't be used as ideal genetic carrier in clinical practice because of their respective defect. But the genetic carrier is key point to gene therapy, consequently many researchers spare no effort to find more safe, high performance and stable genetic carrier. The liposome is more safe and no immunogenicity, but it's efficiency of genetic transmission is lower compare with viral vector. Therefore, study about liposome is main centered to improve the efficiency of genetic transmission. The liposome coupled with antibody improves the efficiency of genetic transmission by the targeted specificity of antibody to make the liposome gathered in some tissue or organ.Objective 1. To explore a way to make the liposome to be a perfect gene transfer vector in brain . 2. To improve the specificity and efficiency of gene transfection in the gene therapy using the monoclonal antibody(anti-Lex/SSEA-1) -mediated targeting liposome, and the monoclonal antibody is targeted to the antigen (Lex/SSEA-1) on the surface of the neural stem cells.Methods 1.
