Studies On Ribavirin Sustained-Release Pellets

Ribavirin,an antiviral agent of nucleoside analog, has wide efficacy against virus and restrains lots of DNA and RNA copy. Ribavirin has stronger activity , wider applications in the clinic and less side-effects .It has been widely used clinically to restrain virus hepatitis , parotitis , infection in upper aspiratory path, flu and SARS. Nowadays Ribavirin is taken 3-4 times daily and shows bigger fluctuation in the drug concentration. The aim of this work was designed to develop a kind of Ribavirin sustained-release pellets,which would reduce the dosing frequency, narrow the blood concentration fluctuation and be more convenient for patients to use.UV method was developed for assaying the drug release from Ribavirin sustained-release pellets. HPLC method was utilized to determine the Ribavirin content in sustained-release pellets . Ribavirin is freely soluble in water or various physiological pH fluids and the apparent partition coefficient in n-octanol/water system is 0.055.To investigate the absorption characters of Ribavirin in the intestine, the absorption kinetics and the absorption sites were studied by utilizing the rat intestinal absorption recirculating method in situ.
