Nicotinic acid belongs to vitamin B, it can inhibit the release of free fat acid in the fat tissue , so it can reduce the total amount of free fat acid arriving liver utilized to synthesize the triglyceride ; the Nicotinic acid can interfere the synthesis of triglyceride in liver and furtherly reduce the form of triglyceride. Different from other effective drug used to decrease fat , Nicotinic acid has many other advantages , it can reduce 40 percent of lipoprotein and increase 40 percent HDL-C .Up to date , it is known the only compound which can make the basic composition of fat in blood normal .But three times a day administration is inconvenient for patients and its liver toxicity is severer. The purpose of the present study was designed to develop a kind of the Nicotinic acid sustained release pellets which would decrease adverse effects and improve its bioavailability, and which would be more convenient for patients to use.Ultraviolet spetrophotometry method was utilized to determine the content in the pellets and assay drug release from Nicotinic acid sustained released pellets, it was adopted to control the quality in vitro by methodology validation.The preformulation results showed that Nicotinic acid is sparingly soluble in water , saline and various physiological pH fluids, the octanol/water partition coefficients is 0.4055, which has no much change in various physiological pH fluidsNicotinic acid pellets were prepared in a centrifugal granulator by powder layer technology with MCC and lactose as dilute agent and 5% HPMC solution as adhesive agent.
