Ketoprofen was developed by frenchman in 1967 as a kind of nonsteriodal anti-inflammatory analgesic and antifebrile drug. It is high efficiency and low toxicity. It's clinic effect is superior to the same kind of profen. In clinic, ketoprofen is mainly used for rheumatoid, arthritics, rheumatic arthritics ankylosing, gout and dysmenorrhea and so on. In recent decades, ketofrofen has been made into several forms of prepared drug using in clinic, such as capsule, tablet, injection, suppository, ointment and lotion in USA, Japan, Germany, Canada and Danmark. It was in 1982 that china composed ketoprofen also, which has been used in clinic in the forms of tablet and capsule.Because of high extent of the fluctuation of plasma concentration in common preparation, in order to reduce the fluctuation, frequency of using drug and side effects, enhance the safety, efficiency and bioavailability, this experiment will make it into a kind of osmotic pump controlled-release tablet. We study it from three aspects, they are:( I preparation of ketoprofen solid dispersion, (II)Study on arts of ketoprofen monolithic osmotic pump, (III) Study on pharmacokinetics of ketoprofen monolithic osmotic pump.( I) preparation of ketoprofen solid dispersionPrepare ketoprofen-polyrinylpyrrolidone (KPF-PVP) solid dispersion which method of dissolvent. Measuring the dissolution of the different proportion of ketoprofen and polyrinylpyrrolidone in defferent periods of...
