Immunotoxins are now being developed to be a new method for cancer therapy. They are designed to allow the antibody fragment to bind to a cell surface antigen or recepter, while the toxin portion mediates cell death.CEA (carcinoembryonic antigen) molecule is a glycoprotein locating on the surface of malignant cells or in the blood of patients suffering from some kinds of cancer. So it is suggested as a tumor associated antigen (TSA). Anti-CEA antibodies are often used as " homing devices", with improved specificity, delivering cytotoxic radionuclides, chemotherapeutic agent, toxins or prodruges, to kill CEA-bearing tumor cells in vivo. We constructed M-CEA-scFv single-chain antibody by conjuncting the VH and VL AA sequence of anti-CEA monoclonal antibody with repeated ligating peptide (G4S)3 referring to the reported method.PE (Pseudomonas exotoxin A) is a 66 kDa polypeptide possessing three domains.
