Gegen Qinlian pellets (GGQL pellets), listed in first section of the 2000s' edition of Pharmacopoeia of the People's Republic of China, are composed of the extracts of Radix Puerariae,Radix Scutellariae baicalensis, Rhizoma Contidis and Radis Glycyrrhizae. It is primarily applied to the treatment of diarrhea, dysentery and enteritis. In this thesis ,GGQL recipe was selected as the model drug of traditional Chinese medicinal compound recipe to treat chronic Ulcerative Colitis because of the simple recipe, more extensive basic researches and good curative effects. The indexes including chemical composition and biological activity, were adopted to study the extracting technology of active fraction of medicinal materials in prescription. The qualitative and quantitative analysis methods were developed to control the inherent quality of formulation. Based on these results, the pH, time and enzyme controlled pulsatile capsule with Gegen Qinlian pills delivery to the colon was investigated.The methods of UV were developed to determine the contents of Pueraia Flavonid, total Baicalin, Berberine alkaloid and total glycyrrhetinic acid; and HPLC for Puerarin, Baicalin, Berberine Hydrochloride and Ammonium Glycyrrhizinate respectively. Above analytical methods were simple, accurate and suitable for the future study of GGQL recipe.The extracting technology of four herbs in the recipe were set up respectively. The total puerarin flavone (TPF) was extracted from Radix Puerariae with 70% alcohol and purifyied with macropore resin 101 and the average content was 84.1%. Chitosan flocculation was used to clarify Radix Scutellariae baicalensis decoction and the optimized conditions were 1% chitosan flocculation agent, 40℃ and pH6.
