Evratamia hainanensis Tsiang is distributed in the Hainan, Guangdong, Guangxi and Yunnan provinces of China. The roots of this plant have been used in Chinese folk medicine for the treatment of stomach-ache, dysentery, snake-bites, rheumatic arthritis, hypertension and virus hepatitis. Because of the extensive medicine in folks, the plant of Evratamia take the attention of the international scholar, for example, the Evratamia officinalis have been used in Chinese folk medicine for the treatment of stomach-ache, sore throat, Evratamia divaricata have been used in Chinese folk medicine for the treatment of opthalmopathy, scabies and pain of breasts, Evratamia coronaria has been used as a cancer remedy in Taiwan, Evratamia polyneura is used by natives in the treatment of ulcerations and Evratamia corymbosa are used in traditional medicine for treatment of syphilic and orchitis ulcerations in Malasia. There was more than 60 species of genus Evratamia in the world, which have been reseached. The number of indole alkaloids was reported to be about 250. Evratamia comprises a large family of 15 species and 5 varieties in our country. Hitherto, there are few chemical constituents or bioactivities of E. hainanensis have been reported. In order to research the bioactivities of E. hainanensis, we have studied the constituents of E. hainanensis. In the course of our search for active components from this plant, the dried roots (5.5kg) of Ervatamia hainanensis TSIANG were exhaustively extracted with 95% EtOH at refluxed temperature. Then the EtOH extract was concentrated under reduced pressure to give a residue (180g), which was suspended in H2O, extracted successively with EtOAc and n-BuOH. The combined EtOAc layers were evaporated to dryness in vacua to give a residue (82g), the residue was subjected to column chromatography on silica gel using petroleum ether-acetone (in gradient) as the eluting solvent, followed by MeOH to yield 8 fractions (Fr. 1-8). Fr.1 provided white powder 3 which upon recrystallization. Compound 5 was crystallized from Fr.2 as colorless needles. Fr.3 were rechromatographed on silica gel using petroleum ether-EtOAc (in gradient) as the eluting solvent to yield 2 subfractions (subfr. 1-2), compound 7 was crystallized from Fr.3.1 as colorless needles, compound 2 was crystallized from Fr.3.2 as colorless crystals.
