Investigation on the optimization of extraction technology, isolation and purification, determination of the chemical structure, anti-tumor activity of polysaccharides from Spirulina platensis(psp) was carried out in this paper. The orthogonal design was employed to optimize TCA method, and the final result indicated that the optimum pH of water, temperature and time of extraction was 12, 90℃ and 8h, under this optimum condition the polysaccharides gained from Spirulina platensis was white powder, the total carbohydrate contents and protein contents was 81.79% and 0.57%. The optimized TCA method was significantly better than the traditional TCA method and more convenient, rapid and economical than the traditional Sevag method. Superdex-75 gel filtration chromatography was employed to isolate and purify crude psp and four polysaccharide compositions with different molecular weights psp1,psp2,psp3 and psp4 were prepared, their molecular weights tested were 65400,16900,14400 and 11700, and the psp2 polysaccharide composition was the main component of psp in the proportion of 80%. Meanwhile psp2 was decompounded through microwave decompounding method and psp5 polysaccharide composition with lower molecular weight (8900) was gained. Gel filtration chromatography, Gas chromatography, Infrared spectrogram, Nuclear magnetic resonance(NMR), NaIO4 oxidation, Sulfuric acid-carbazole method and Barium sulfate turbidity method were employed to investigate on the primary chemical structure of psp2.
