Objective: To evaluate the protective effect and explore the protective mechanism of Huqiyin on bleomycin-induced pulmonary fibrosis in rats.Methods: The SD rats were randomly divided into six groups, including normal group (Group A), control group (Group B), prednisone group (Group C), low-dose group of Huqiyin (Group D), medium-dose group of Huqiyin (Group E) and high-dose group of Huqiyin (Group F). The pulmonary fibrosis model was established by a single intratracheal injection of 5mg/kg of bleomycin. The rats of Groups C, D, E and F were treated with corresponding medicine. On the 28th day, rats were sacrificed under anaesthesia, and serum and lung tissues were collected. The serum and left lungs were used for hydroxyproline (HYP) analysis, and the right lungs for histopathological studies (HE and VG staining). The expression of transforming growth factor-β1 (TGF-β1) protein and mRNA in lungs were assessed by immunohistochemistry (IHC), in situ hybridization (ISH), respectively, and analyzed with quantitative image system.Results: 1. The weight gain of rats in Groups E and F were higher than that in Group C (P<0.05) at the 7th, 14th day, but no significant difference were observed compared with Group A.2. The scores of alveolitis and pulmonary fibrosis of Group B were greater than ones in Group A (P<0.01), and those of Groups C, E and F were lower than ones in Group B (P<0.01).
