The primary factor to restrict the efficiency of antitumor drugs is its serious side effects and the resistance of tumor cells against these drugs. While the antitumor drugs repress or kill tumor cells, the normal tissues and cells would also be involved without escaping damage by luck, which will result in a lot of toxic impacts. In addition, the drug-resistance, developed during the process of drug-usage, is another essential factor to limit the clinical efficacy of chemotherapy. The mechanisms involved in the drug-resistibility by tumor cells are multiple, the multidrug resistance(MDR), however, is the high-light up to date, which functions by active drug efflux through membrane transporters such as P-glycoprotein (P-gp) and multidrug resistance-associated protein (MRP). The changes in drug targets (receptors, enzymes and so on), the upregulation of salvage anabolic pathways and DNA repair mechanisms etc are all closely related to drug-resistibility. But many experiments using MDR modulators for reversing the drug-resistibility have not yet produced promising results.
