Objective: To investigate the activity change of Caspase 1 and Caspase 8, apoptosis of PMN in acute pancreatitis of rats and their relationship and the possible mechanisms involved.Methods: 90 rats were randomly devided into three group(ANP group, AEP group and the control group), each only 30. AP model was separately developed in adult rats by retrograde injection of 3%(ANP group) and 0.5%(AEP group) sodium taurocholate in the pancreatic duct. The control group only underwent pancreatic duct cannulation without sodium taurocholate. At the time of 0,3,6,12,24h,get the pancreas for pathobiology and immunohistochemistry,get the blood from inferior vena cava and separated PMN,detected the apoptosis of PMN by flow cytomery,measured the activity of Caspase 1 and Caspase 8 in PMN by fluorescent quantitative assay,detected the activity of Caspase 1 and Caspase 8 in pancreas by immunohistochemistry.Results: With prolongation of time, the pancreatitis developed stepwisely, apoptosis of PMN in ANP group was delayed markedly while activated in AEP group. At 24h,the apoptosis index of PMN in ANP decreased to 1.87± 0.41, while it in AEP increased to 9.4±0.33 at 6h.
