Objective:We successfully induced the acute pancreatitis models by injecting directly 5% sodium taurocholate solution into common biliopancreatic duct of the Sprague Dawley rat.With the SD rats' extension of survival time,whether the severity of acute pancreatitis was associated with survival time? How did the protein of the c FLIP and caspase-8 related to cell apoptosis and necrosis expressive degree? And whether the two proteins was connected with the severity of acute pancreatitis? Methods:We built the models of acute pancreatitis by retrograde infusion of 5% sodium taurocholate solution into common biliopancreatic duct of the Sprague Dawley rat with the 24 G intravenous detained needle.We set five groups: the experimental four groups giving sodium taurocholate solution that is the three-hour group,the six-hour group,the twelve-hour group,the twenty four-hour group,and the twelve-hour control group without giving any chemical agent.We dissected rats in the condition of intraperitoneal anesthesia after the successful construction of the rat models and observed whether the pancreatic tissue,pancreatic surrounding tissue,mesentery and greater omentum had calcium saponification and whether the abdomen had ascites and the change of the quantity and quality of ascites.We calculated the weight of ascites with the weighing method(the weight of ascites =wet cotton ball weight-dry cotton ball weight)(g).We got the pathological score of pancreatic tissue through the HE staining and observed the pathomorphological changes of pancreatic tissue under microscope.We analyzed the pancreatic tissue protein of the cFLIP and caspase-8 expressive level using Western blot technique.Results:The Sprague Dawley rat models of acute pancreatitis all had reached the scheduled experiment time in the condition of free drinking.According to the different requirement of the rat's models,we dissected rats in the condition of intraperitoneal anesthesia and observed the change of ascites.With the extension of survival time of the animal models of acute pancreatitis,the weight of ascites was increasing and the color of ascites was deepened gradually.And we also observed the pancreatic tissue,pancreatic surrounding tissue,mesentery and greater omentum of the twelve-hour and twenty-four-hour experimental group had different pathological damage.With the extension of survival time of the Sprague Dawley rats,the degree of pancreatic tissue damage increasing gradually.The result of the control group VS the three-hour experimental group VS the six-hour experimental group VS the twelve-hour experimental group VS the twenty four-hour experimental group is(edema: 0.25±0.27 VS 2.17±0.26 VS 2.58±0.20 VS 2.92±0.38 VS 3.92±0.20;pancreatic acini necrosis: 0 VS 0.50±0.00 VS 0.67±0.26 VS 1.58±0.38 VS 3.25±0.52;bleeding and fat necrosis: 0 VS 0.58±0.20 VS 1.00±0.32 VS 1.92±0.38 VS 2.50±0.00;inflammatory cell infiltration: 0 VS 0.58±0.20 VS 1.08±0.20 VS 1.75±0.27 1.75±0.27 VS 2.83±0.26).With the models' extension of survival time of acute pancreatitis,the protein expression level of the caspase-8 decreased gradually.And the result of the three-hour experimental group VS the six-hour experimental group VS the twelve-hour experimental group VS the twenty four-hour experimental group is(592.20 VS 457.82 VS 387.50 VS 363.50).The protein expression level of the cFLIP-S contrary to the caspase-8 and the result of the three-hour experimental group VS the six-hour experimental group VS the twelve-hour experimental group VS the twenty four-hour experimental group is(430.37 VS 493.49 VS 545.58 VS 595.97).Conclusion:With the extension of survival time of the acute pancreatitis models of the Dawley rats,the necrotic degree of pancreatic tissue was increasing gradually,but the apoptotic degree of the pancreatic cell was relieved.The expressive level of the Caspase-8 was positively correlated with the pancreatic parenchyma cell apoptosis,and negatively correlated with the cell necrosis.And the cFLIP-S was positively correlated with the pancreatic parenchyma cell necrosis,and negatively correlated with the cell apoptosis.The caspase-8,cFLIP-S will be able to become the laboratory examination indexes of the early clinical evaluation of the severity of illness,the effect of diagnosis and treatment,and clinical prognosis in patients who suffer from acute pancreatitis in future.
