Objictive: To investigate the expression of hypoxia-inducible factor-1 alpha(HIF-1α) and cyclooxygenase-2(COX-2) protein in middle ear cholesteatoma epithelium. and discuss their action in cholesteatoma.Methods: we studied 36 cases of middle ear cholesteatoma epithelium who were operated in First Affliliated Hospital of Guangxi Medical University, and compared with 20 cases of external auditory canal epithelium. All of them had been verified to be cholesteatoma by pathology. The expression of HIF-1αand COX-2 protein were detected by immunohistochemical method.Results: The result showed that HIF-1αand COX-2 could be detected in all middle ear cholesteatoma epithelium but there were almost no positive cells in external auditory canal epithelium. HIF-1αwas located at karyon, taking on yellow-brown pellet, COX-2 was located at cytoplasmic, taking on yellow-brown pellet too. In cholesteatoma epithelium samples, HIF-1αpositive rate was 88.9%, and COX-2 positive rate was 94.4%. Compared with external auditory canal epithelium, the espression of HIF-1αand COX-2 increased in middle ear cholesteatoma epithelium (P<0.05). The expression of COX-2 were significantly increased in extensive cholesteatoma in comparison with localized cholesteatoma, P<0.05. HIF-1αand COX-2 didn't have significant correlated by Spearman test.
