PARTⅠExpression and Correlation of HIF–lαand COX-2 in PsoriaticLesionsObjective: To study the expressions and Correlation of HIF-1αandCOX-2 in samples of psoriasis vulgaris tissue and those of skin tissue whichwere from healthy individuals. To explore the role of HIF-1αand COX-2 inthe pathogenesis of psoriasis .Methods: 30 patients with psoriasis vulgaris and 20 normal humancontrols were enrolled into the study and obtained these lesions or normalskin tissues(1)RT-PCR was used to examine the mRNA expressions ofHIF-1αand COX-2 in samples of psoriasis vulgaris lesion tissues and those ofskin tissues from healthy individuals。(2)Western blot was used to examinethe protein expressions of HIF-1αand COX-2 in samples of psoriasis vulgarislesion tissues and those of skin tissues from healthy individuals。Results: The expressions of HIF-1αand COX-2 mRNA in psoriasisvulgaris tissues(1.32±0.04 and 1.21±0.04)were significantly higher thanthose in the normal group (0.49±0.03 and 0.31±0.03). Which showedsignificant difference in the expressions of HIF-1αand COX-2 mRNAbetween the psoriasis and the control group(P<0.05).Moreover, The HIF-1αmRNA in psoriatic lesions was associated significantly with COX-2mRNA(r=0.664,P<0.05). The expressions of HIF-1αand COX-2 protein inpsoriasis vulgaris tissues(0.76±0.03 and 0.62±0.03)was significantly higherthan those in the normal group (0.44±0.05 and 0.28±0.06). which showed significant different in the expressions of HIF-1αand COX-2 protein betweenthe psoriasis and the control group(P<0.05);And HIF-1αprotein in psoriaticlesions was associated significantly with COX-2 protein (r=0.601,P<0.05)。Conclusion: HIF-1αand COX-2 high expression and their positivecorrelation in psoriatic lesions might play an important role in genses anddevelopment of psoriasis. PARTⅡExpressions and Correlation of HIF-1αand COX-2 inepidermis of guinea pig psoriasis-like animal modelObjective: To detect HIF-1αand COX-2 expression in epidermis ofguinea pig psoriasis-like animal model and their correlation and role in thepathogenesis of psoriasis.Methods: Psoriasis-like animal model was established by topicaltreatment with 5% propranolol emulsion on ear’s back of guinea pigs. Wedetected HIF-1αand Cox-2 in normal epidermal tissues from 10 normalguinea pigs and the lesions of ear’s back from 20 guinea pigs psoriasis-likeanimal models by immunohistochemistry.Results: Expressions of HIF-1αand COX-2 in lesions of treated groupwere significandy increased; While there was no positive staining expressionin normal epidermis. Compared with the control group, the positive rate and the mean expression level of HIF-1αand Cox-2 in epidermis of guinea pigpsoriasis-like animal modesl were significantly higher than the control group(P<0.01).Conclusion: The expressions of HIF-1αand Cox-2 in the epidermis ofpatients with psoriasis vulgaris and seborrheic keratosis was higher thannormal epidermal.it has high concordance with human psoriatic lesions notonly in histopathology, but at molecular level.
