Radix Morindae officinalis, one of the valuble Chinese herbs in South China, is a common Chinese medicine included in the 2005's Pharmacopoeia of People's Republic of China, and it origin from family Rubiacae, Morinda officinalis How.. In Traditional Chinese Medicine(TCM), it is used as reinforcing the kidney yang, to strengthening the tendons and the bones, and relieving rheumatic condition, which is used in impotence, seminal emission, infertility, menstrual disorders, pain and cold feeling in the lower abdomen;rheumatic arthralgia;limpness of the legs.The resource of Morinda officinalis How. is distributed in Guangdong Province, Guangxi Province, Fujian Province, and Hainan Province naturally. Guangdong Province is primary region of genuine Radix Morindae officinalis.Section one Literature studyWe looked the weak link and the problem among the studies of the Radix Morindae officinalis, and raised the study content and the object after consulting many studies about Radix Morindae officinalis.Section two Experimental studyChapter one Resource investigation of Morinda officinalis How.The result: the planting area in Guangdong Province is maximal. The germplasm of Morinda officinalis How. have Guangdong Province, Guangxi Province, and Hainan Province.Chapter two Study on quality control method of Radix Morindae officinalis Experiment one: Study on the fast preparation of monotropein Monotropein had been distilled and separated from the leaves of Morinda officinalis How., which avoided the disturbance of polysaccharide.Experiment two: Assay monotropeinObject: To establish an extraction method of monotropein from Radix morindae officinalis;To realize the contents of monotropein in Radix Morindae officinalis and other samples.Methods: The method of HPLC was used with Chromatographic conditions: Kromasil C18 (150mm × 4.6mm, 5μm) column. the mobile phase:(1)extraction method:methanol-0.4% phosphate solution(10:90),(2)assay the contents of monotropein:methanol-0.4% phosphate solution (5: 95→28.8: 71.2, 15min) .the velocity of flow was 1mL · min-1;the detection wavelength was at 210nm;and columntemperature was 25℃.Results and Conclusion:1. The ultrasonic extraction with 80% MeOH as much as two times of the medicine amount for 30min and twice was more effective, The kinds of solvent and models of extraction have a great influence upon the extracting efficiency of monotropein.2. The effect of different growth stages of Morinda officinalis on monotropein. The contents of monotropein from the 2.5,3-5,4.5,5.5 growth years were (7.68±0.04) mg ? g"\ (13.29 + 0.14) mg ? g\ (13.90 + 0.10) mg ? g"\ (16.47 + 0.45) mg ? g"1, respectively. It is the best collecting stages when grow four years.3. Assay the contents of monotropein in Radix Morindae officinalis from different producing areas.Assay the contents of monotropein in 70 individual from 4 kinds of different producing areas. The contents of monotropein in Radix Morindae officinalis from different producing areas are different obviously.4. Assay monotropein in Radix Morindae from different processed products There were differences between the crude and samples of different processedproducts of monotropein in the Radix Morindae officinalis, the contents of monotropein from the Different Processed Products (crude drugs without wood, samples removed from wood, samples processed with salt, preipared samples) were 13.92 mg ?g"\ 9.10 mg -g\ 9.21 mg *g"\ 12.86mg 'g'1 respectively. Comparison of the HPLC profiles of total idridoid of Morinda officinalis How. and its adulterants, the results indicated that they have distinct differences each other, the similarity assessment of quality goods is more than 0.94, which of mostly adulterants is less than it. We can utilize the similarity assessment to distinguish the quality goods and its adulterants accordingly.3. The HPLC chromatogram which corned from different habitats is distinct, by which We can distinguish the different habitat's products.4. The HPLC chromatogram is quite a difference of different processed products, which indicated that the different processing methods could change the iridoid component.5. a chart had been stipulated to evaluate the quality of commercial Radix Morindae officinalis, which indicated that the most of samples are unqualified.
