Object:In this experiment ,we observed the local application of IGF-1in the osteoporosis rats, and draw a scientific conclusion through the dynamic state observation to the union of fracture regional, aimed to provid rational and according for the further clinical application.Methods: We elected 96 female SD rats of majority, they were randomly divided into normal control group(N), bone rarefaction group(O),and four experiment groups(A,B,C,D), besides the normal control group, remove the double ovavies after 12 weeks, to result in bone rarefaction model. All the rats were formed to fracture of tibia models, and from the first day after the tetechnic, different injection of 0.2ml/100g were used for continuous two days. normal control group(N),bone rarefaction group(O) were injected distilled water; and four experiment groups were injected insulin-like growth factor-1(A 1ng/kg,B10ng/kg ,C100ng/kg ,D1000ng/kg.).We draw samples among each group to do the X-check, by the 12 day, the 21day, the 30 day and the 40 day,the stress observation in fracture of tibia and histology observation. All the data were analyzed by statisticResult:⑴The healing process in normal control group(N), bone rarefaction group(O) accorded with the change rule of normal bone fracture union. It is to say that the quality of union of fracture after bone rarefaction depressed.⑵Imageology: there are manipulus callus formation after 12 days in D group. After 21 days, callus formation in all groups, the fracture line blured in group C and D. After 30 days, there were multitude callus formation, and partly thickening. After 40 days, there were ponderosus callus formation in bone fracture of group C and D, partly thickening, bone density ,the fracture line lured in other groups.⑶Vitodynamics: Among the groups, following the technic time extended, crushing strength gradually enhanced and peak value appeared, that is early and identify in D group.⑷Histology: Among the time after bone fracture ,cartilage cells in all groups multed, the numbers in group D incresed fastest, the next in group C ,and the numbers in group O incresed slowliest. The agglutination in group C and D ahead of other groups,and group D is better. All these are corresponded with that in imageology. Conclusion:1)The partly injection of IGF-1 may promote the union of fracture of bone rarefaction. 2)The degree of IGF-1 promoting the union together with the injected dose and dose-dependent.
