| Objective To investigate the effects of PGE1(prostaglandin E1)on the expressions of transforming growth factor-β1 (TGF-β1) and connective tissue growth factor (CTGF)in renal interstitium in rats with obstructive nephropathy.Methods A total of 60 Wistar rats were divided into experiment group (include PGE1 treatment group , and group with normal saline) and control group (sham operation group) . Models of rats with obstructive nephropathy were established by unilateral ureteral ligation. The rats were sacrificed at3, 7 , 14 , and 21 day after operation and specimens were taken from the serum and renal cortex. The morphopathological changes in the renal interstitium were observed by light microscopy. CTGF , and TGF-β1were detected by immunohistochemical staining. The measured data were evaluated by Mais-2000 image analysis system.Results 1. In 3,7 day, the renal index of PGE1treat group were higher than sham operation group(P<0.05).Comparated with the sham operation group ,the saline group's renal index were higher in 3,7,14day (P<0.05). PGE1 treat group were lower than saline group 7 and 14day. 2 Masson staining shows that the experiment group's fibrosis was more serious than sham operation group's fibrosis (P<0.05). The fibrosis of PGE1 treat group were lower than that of saline group (P<0.05).3.Semi-quantitative analysis indicated that the expression levels of CTGF and TGF-β1 in the renal interstitium in the sham operation group were significantly lower than those in the experiment group at 3,7,14,21d after operation (P<0.05).4.The expression of TGF-β1 and CTGF in the renal was closely correlated with renal interstitium fibrosis(r=0.835, P﹤0.05; r=0.803,P﹤0.05). Conclusion PGE1 may inhibit renal interstitial fibrosis in rats with obstructive nephropathy by down regulating the expressions of CTGF and TGF-β1 in the rat renal interstitium. |
