Objective:To discuss the feasibility of osteonecrosis and bone defect of the rabbit femoral head treated with vascular bundle implantation associated with allogenic bone marrow stromal cells (BMSCs)transplantation. To explore a new theraphy on treatment of osteonecrosis of femoral head, and to establish the experimental foundation for further clinical application.Methods:1 The preparation for allogenic bone marrow stromal cells transplantation:Firstly, the rabbits'posterior iliac crest were punctured and 3ml marrow was withdrawled, which was inoculated in 6-well dishes.Secondly,5ml conditional DMEM medium (15% fetal bovine serum,100u/ml penicillin sodium,100u/ml streptomycin,10mmol/Lβ-glycerophosphate sodium,10-8mmol/L dexamethasone,50mg/L vitamin C) was mixed into it,and then the 6-well dishes was put into humidified incubator. The medium was replaced for the first one after 5 days, since then,it was replaced every 3 days.Cells of the 3rd generation were kept refrigerated in liquid nitrogen, and were revived after 7 days. When cells of the fifth generation were born, 100μl BrdU was mixed into it,and the telo concentration of...
