Valeriana amurensis is mainly distributed in the Northeast of China and abundant in resource in the Greater Xingan Mountain Region. So far, there is no research work published in V. amurensis although some relative plants in the same genus have been widely used to cure insomnia and other diseases.In this thesis, the morphology of original plant, exterior characters and microscopic features of the dried root and rhizome of Valeriana amurensis were studied and will provide an academic basis for the identification of Valeriana amurensis.A optical extracting method was determined by comparing three extractive methods of steam distillation, isolated distillation and supercritical CO2 extraction, according to the extractive ratioes of the volatile oil and the contents of sedative components Valeriana amurensis. The components of the volatile oil extracted from Valeriana amurensis were analysed with GC-MS, and the physicochemical property of the volatile oil was also determined.Prominent CNS inhibition effects of the volatile oil extracted from Valeriana amurensis was proved by pharmacological investigation, including the experiments of autonomic activities, synergism with barbiturates drugs, writhing caused by acetic acid and antagonize hyperspasmia induced by thiosemicarbazide of mouse. Besides, the sedative mechanism of the volatile oil from Valeriana amurensis was investigated by determinies the influence of it to the electroencephalograms of normal rats. The research results set an academic foundation for develop new drugs of improving sleep quality.
