Isolation And Identification Of Glycosides From Forsythia Suspense Leaves And Study On Its Anti-fatigue Effects

This text compared the anti-fatigue effects between two kinds of the FSG that prepared from Forsythia Suspense leaves and identified by HPLC, so as to provide some evidences for the utilization of forsythia suspense leaves which has not been exploited up to now.The glycosides of Forsythia Suspense leaves such as forsythin and forsythoside are effective components. This experiment firstly prepared the glycosides from the Forsythia Suspense leaves by column chromatography of macroporous adsorption resin, then the content of forsythin and forsythoside in four products were assaied by HPLC, finally chose two glycosides which had more effective components than others to do the anti-fatigue experiment.In the anti-fatigue experiment, firstly the animal model of exhaustive swimming exercise was made in mice, then SOD, MDA and so on were measured to estimate the effects of FSG â…¡ and FSGâ…¢; The expression of CuZn-SOD gene was studied by immunohistochemical method; Electron microscope was used to observe the effect of FSG on ultrastructure in cardiac muscle, liver and quadriceps femoris.The results show:1. The content of forsythin and forsythoside in both FSG â…¡ and FSGâ…¢ are higher than others, so it is inferred that both of them have good effects. The content of forsythin in FSG â…¡ are higher than FSGâ…¢, but FSGâ…¢ are easily to prepared, and have lower prepared cost than FSG â…¡, so FSGâ…¢ have better exploitable value than FSG â…¡.2. FSG â…¡ and FSGâ…¢ can improve the sports capability of mice, prolong the exhaustive time of swimming, and FSG â…¡ have better effects than FSGâ…¢.3. FSG â…¡ and FSGâ…¢ can raise the content of Hb and the activity of CAT in erythocyte, and FSG â…¡ have better effects than FSGâ…¢. Maybe there are something in FSG can stimulate the synthesization of erythocyte.4. FSG â…¡ and FSGâ…¢ can improve the blood sugar and decrease the BUN in blood serum, and FSG â…¡ have better effects than FSGâ…¢.5. FSG â…¡ and FSGâ…¢ can increase the activities of T-SOD and CuZn-SOD, decrease the content of MDA in cardiac muscle, liver and quadriceps femoris. Thus reduce the injure of tissue cell that induced by free radical(FR).
