| Objective:To observe inhibition of growth and angiogenesis of human gastric carcinoma SGC-7901 cell line xenografts in nude mouse model by tea (-)-epigallocatechin-3-gallate (EGCG) as well as investigate molecular mechanism.Methods:1.The effect of tea(-)-epigallocatechin-3-gallate (EGCG) on proliferation of human gastric carcinoma SGC-7901 cell line in vitro: The growth inhibition of SGC-7901 cell was measured by MTT assay, growth curve analysis, double time and vitality detection.2.Establishment and group of human gastric carcinoma SGC-7901 cell line xenografts in nude mouse model : BALB/C mouse were transplanted by ip injection with 2×10~6 SGC-7901 cell respectively. A total of 30 nude mouse were randomly divided into 6 groups: Sodium Chloride group, 5-Fu control group, Minocycline control group, 5mg/kg EGCG group, 10mg/kg EGCG group, 20mg/kg EGCG group. After the tumor formed, Sodium Chloride control group was injected with NS; Control groups and EGCG groups were injected with drag through abdominal cavity, respectively.3. The evaluation of EGCG was detected by human gastric carcinoma SGC-7901 cell line xenografts in nude mouse model.Transplant tumor's weight and volume were observed; The expression of nuclear factor-_KBP65 (NF-_kBP65) protein was observed by western blot in different groups ; The expression of VEGF in xenograft tumor cells was analyzed by indirect immunofluorescence flow cytometry.
