| Objective The present study was declined to screening the high invasivemetastatic strain of histiocytosarcoma L - â…¡ by constant subculturing of the cells from the pulmonary metastasis in vivo. Then its biological behaviour was identificated initially. To compare with two kinds of strain of distinct metastasis ability in vivo and vitro, it could conduce to better understand the molecular mechanism of tumor metastasis and study of anti-tumor metastasis.Methods We subcutaneously injected histiocytosarcoma L - â…¡ cells inthe right flank of mice, then isolate the cells in pulmonary metastasis and injected them in another mouse by the same methods. After several subculturing, we have screening the high invasive metastatic strain of histiocytosarcoma L- â…¡. We analyze the isolated cells by morphlogy analysis(light microscope,electron microscope), cell growth curve, mean life span with cancer, karyotype analysis, flow cytometry and compare with contrast of rate of lung metastasis and immunohistochemistry of two kinds of strain of distinct metastasis ability.Results Morphology observation: There are samg morphous and constructionbetween the 1st and the 7th generation solid tumor under light microscope. The tumor appear polygon, disparity size, bigger karyoplasmic ratio, round, ellipse or anomalism of nucleus, lots of karyokinesis and pathological karyokinesis. The 7th generation solid tumor are more wide intercellular space, abundant mitochondrion and Golgi apparatus than 1st generation under electron microscope. In the surface of 7th generation solid tumor could be discover much microvillus. |
