Objective: To investigate the effect of Mifepristone or Cisplatin or both on human cervical squamous carcinoma cells cultured in vitro,and its mechanicsm. Methods: Human cervical squamous carcinoma Caski cell line nomal controlgroup:Human umbilical vein endothelial ECV-304 cell line and Human cervical adenocarcinomal Hela cell line which express PR(progesterone receptor) protein as PR positive control group were cultured in vitro .Caski cell line was treated with various concentration of RU486 or CDDP or both, ECV-304 cells were treated with various concentration of RU486.Cell proliferation of Caski,ECV-304 cells were evaluated by MTT assay ; The ability of clone formation of Caski cells was evaluated by clone formation test;The apoptosis rate and cell cycle of Caski ,ECV-304 cells were determined by PI staining flow cytometry ;The shape of cell apoptosis of Caski cells were determined by AO/EB double fluorescence stain; The expression and their activity changes of HPV16-E6n p53 Bcl-2 Bax protein of Caski cells were investigated by FITC staining flow cytometry;The expression of PR protein of Caski and Hela cells were investigated by immunohistochemical techniques.Resluts: (1) MTT assay indicated that RU486 which concentrateion excessed12mg/L could inhibit the proliferation of Caski cells in a concentrateion dependent and time dependent positive; while the proliferation of ECV-304 cells was not affected at the same concentration;RU486 at concentrations 1.25 2.5mg/L were not of cytotoxicity to Caski cells (IR<5%), but they significantly increased the cytotoxicity of CDDP to Caski cells ,these results were consistent with the results of Colony formation test on Caski cells. (2) AO/EB double fluorescence stain and PI staining flow cytometry data showed the apoptosis of Caski cells was induced and cell cycle was arrested at G1 phase by RU486 whose concentration excessed 12mg/L in a concentration and time dependent manner ;However, PI staining flow cytometry data...
