ObjectiveTo investigate the effect of CC1007, a novel small-molecule compound, on chemotherapy sensitization of cisplatin for cervical cancer cells and its mechanism.Methods1. Cervical cancer cell lines, HeLa and Caski, were treated with CC1007、cisplatin either as a single agent or in combination for48hours. The cell proliferation inhibition was measured by MTT assay. Calculated the50%inhibiting concentration (IC50) of cells and made the concentration-inhibition rate curve. Experiments were repeated for three times.2. Cervical cancer cell lines, HeLa and Caski, were treated with CC1007、cisplatin either as a single agent or in combination for24hours. The expression of Nur77was measured by western blot. Experiments were repeated for three times.3. Statistical analysis was carried out on the experimental data.Results1. Both of CC1007and cisplatin have a dose-dependent inhibitory effect on cervical cancer cells, and the differences were statistically significant (P<0.05). Cotreatment of CC1007with cisplatin markedly enhanced cell death, and the differences were statistically significant (P<0.05). CC1007and cisplatin inhibit cell proliferation in a synergistic manner. 2. Compared with the control group, cisplatin and cotreatment of CC1007with cisplatin upregulated the expression of Nur77both in HeLa and Caski, and the differences were statistically significant (P<0.05). CC1007upregulated the expression of Nur77stimulated by cisplatin.Conclusions1. Both of CC1007and cisplatin have a dose-dependent inhibitory effect on cervical cancer cells. Cotreatment of CC1007with cisplatin markedly enhanced cell death. CC1007and cisplatin inhibit cell proliferation in a synergistic manner.2. Cisplatin upregulates the expression of Nur77, and the effect is markedly enhanced in combined with CC1007, which may be the mechanism of inhibiting the cell lines in a synergistic manner.
