Background and AimsIt was thought that the dead cardiocytes can only be repaired by the formation of scar organization, and the medicine can be used to defer the cardiac remodeling to lower the occurrence of the congestive heart failure and the mortality of the clinic. In recent years, people always look for a kind of cells to replace the dead cadiocyte to maintain the normal heart function, and find that the stem cells transplantion bring the new hope to the treatment of myocardial infarction.Marrow mesenchymal stem cells(MSCs) are a kind of cells which have the ability of self-duplication, can differentiate into various kinds of cells,and won't be limited to ethics problem. Currently MSCs are regared as the most hopeful stem cell to apply for the substitute treatment of infarction cadiocytes. MSCs were serial passaged for 4 months by Makino, and the immortalized cells were got . By the treatment of 5-aza, part of those cells were differentiated into the cardiocytes-like cells resemble to fetal cadiocytes, which has supported the theory foundation of use of MSCs for the cellular transplantion into the heart. In recent years, MSCs have been studied on how to be induced into cadiocytes under all kinds of conditions, and the identification of the differentiated MSCs mainly focus on the aspects of morphology and immunohistochemistry. Though the study on the electrophysiological characteristics is rare, it's very important that whether the differentiated MSCs possess the electrophysiological characteristics similar to the cadiocyte, which would directly determine that the cells can concert with the host cadiocytes in the function or not, such as excitability, conductivity, etc. Our experiments aim to differentiate MSCs into cardiocytes-like cells by 5-aza in vitro and investigate the membrane currents of the cardiocytes-like cells with the whole-cell patch-clamp technique.Methods1.
