Objective: The type 1 insulin-like growth factor receptor (IGF-1R) belongs to a family of transmembrane protein tyrosine kinases. It is overexpressed in various tumors involve hepatocellular carcinoma and related tightly to progression, invasion and metastasis of tumor. IGF-1R antisense oligdeoxynucleotides(ASODN) can down regulate the expression of IGF-1R , inhibit tumor growth and delay tumor recurrence after operation. The aim of the study was to investigate the therapeutic effect and side effect in treating human hepatocellular carcinoma in nude mice orthotopic implantation model with IGF-IR antisense phosophorothioate oligodeoxynuclieotides and to provide reliable evidence for early clinical use.Methods: Forty cases of human hepatocellular carcinoma in nude mice orthotopic implantation model were established and divided into 5 groups randomly, eight in each group. (1)NS control group, (2)IGF-1R APSODN(75mg/kg) group, (3)IGF-1R APSODN(50mg/kg) group , (4)IGF-1R APSODN (25mg/kg) group and(5)5-FU control group. 72 hours later, the agents were given. It summed to 25 times. The weight of nude mice was measured every 7th day in the course of treatment. When the experiment was over, we begin to analyse the nudes blood cell, measure the volume and weight of tumor, calculate the inhibitory rate of tumor growth, examine the pathomorphological change. This experiment was repeated another time.
