Type 2 diabetes mellitus (T2DM) is one of the metabolic diseases, which Characterized by high blood glucose levels. The classⅡtransmembrance glycoprotein (PC-1) is a potential candidate gene for insulin resistance and T2DM because it inhibits insulin receptor tyrosine kinase activity. The K121Q polymorphism in exon 4 of the PC-1 gene has been associated with insulin resistance and T2DM in many, but not in all studies.Objective To investigate the association between the K121Q polymorphism of the PC-1 gene and obesity and T2DM.Methods 119 patients with T2DM(63 men and 56 women, mean age 58±11.3 yrs) and 442 non-diabetic subjects with fasting glucose less than 5.6 mmol/L or OGTT glucose less than 7.8 mmol/L(227 men and 195 women, mean age 55±10.4 yrs) were selected. Genomic DNA was isolated from peripheral blood cell according to standard procedures. Sense and anti-sense primers were designed according to the sequence of PC-1, 238bp fragments of polymerase chain reaction spanning the K121Q nucleotide was amplified. The produce were digested with restriction enzyme AvaⅡfor twelve hours.The genetypes were read after separation of the digested products with 8% SDS-PAGE. Differents distribution in genetypes between T2DM and non-diabetetic subjects was analyzed and between obesity and non-obesity in Dalian Han nationality and between diabetes relative and non-diabetes relative in diabetic patients.
