Objective : The pathogenesis of the ulcerative colitis(UC) is complex. The definite aetiology of UC remains unknown. Recent observations indicated implication of peroxisome proliferator-activated receptors (PPARs) in the inflammatory response and inflammation-related disorders as well as in the control of metabolism of the lipid and lipoprotein, especially PPAR-γ. We examined the expression of PPAR-y in the biopsy of colonic mucosa of patients with ulcerative colitis(UC) and assessed its relationship with the seriousness of disease .Methods: The biopsy specimens were taken endoscopicaily from 25 patients with ulcerative colitis (UC) and from 15 control separately. The expression of PPAR-y antigen in colonic mucosa was detected by immunohistochemisty of the labeled-avidin-biotin method .The first antibody was rabbit to human monoclonal IgG antibody against PPAR-γ and 0.01MPBS take the place of the first antibody to be the blank control. The positive cells are dyed brown. Ten high power field are detected from every specimen randomly then calculate the percentage of the positive cells.Results: PPAR-y expressed chiefly in the nuclear of the mucosa cells. The percentage of positive cells in the lesion of UC and its juxta-mucosa and control were 22.8% ±8.9%, 29.00 %±5.7%, 38.00% ± 6.7%, respectively (P<0.05 ). The expression in UC was lower than that in control. The positive expression in mild, moderate, severe degree of UC were 29.5%±7.2%, 22.0%±4.8%, 11.00%±4.1%, respectively (P<0.05) . The...
