CCR5 is the most important co-receptor in the early stage of HIV infection.Recently, the strategy targeting CCR5 to block HIV infection have been proved effective and useful. Several of approaches such as CCR5 antagon, CCR5 mAb and RNAi which can attenuate the function of CCR5 or lower the amount of CCR5 in the cell membrane can cut down the HIV infection rate.CCR5 combined autovaccine strategy developed a new way to the AIDS vaccine research. Moreover, CCR5 autovaccine may induce anti-CCR5 antibodies that can bind native CCR5 and block viral infection in vivo. In our work, four CCR5 combined autovaccine antigens were constructed and expressed in the prokaryotic expression system. After that we have developed different approaches to purify the four recombinant protein.The purified protein were immunized into the animal and anti-serum were collected.The further characterization of anti-serum immunological activity were investigated.
