| To elucidate the relationship between the underlying mechanism of penicillins allergy and food, aeroallergens hypersensitivity, radioallergosorbent (RAST) was used to examine 8 kinds of specific IgE antibodies of different antigenic determinants of penicillins (BPO, PVO, APO, AXO, BPA, PVA, APA and AXA) and 8 kinds of specific IgE antibodies of common antigens (including peanut, milk egg, crab, house dust, dust mite, spring pollen, ragweed pollen) in 219 patients with penicillins allergy and 121 patients with food and/or aeroallergens hypersensitivity. Material and methods1. Patients There are 219 patients with penicillins allergy and 121 patients with non-allergic penicillins allergy (including 38 subjects with food allergy, 68 subjects with aeroallergens hypersensitivity, and 15 subjects with both).2. Determination of specific IgE antibody Radioallergosorbent (RAST) was used to determine the specific IgE antibodies in sera of patients. Antigen discs were incubated 3 hours with undiluted serum at constant room temperature in the tubes. After washing 3 times with phosphate buffer saline (PBS), discs were incubated overnight with 125I-labelled sheep anti-human IgE. After washing 3 times with PBS, bound 125I was determined in a γ-counter.3.
