Objective:To explore whether calcitonin gene-related peptide(CGRP) influence the proliferation and transformation of phenotype of vascular smooth muscle cells(VSMC) and to evaluate whether the organic and cellular models established by cultured aorta of rats are applied to use,the aorta segments and VSMC from cultured aorta of rat were incubated in culture flask containing CGRP DMEM culture medium. The proliferous cells were labeled by Bromodeoxyuridine(BrdU) with immunocytochemical method,and the mRNA expressions of hypertension-related gene-1(HRG-1) and smooth muscle 22 alpha(SM22α) were detected by RT-PCR.Methods:The first experiment (used the organic model of VSMC proliferation): 4 male rats weight between 150-200g were anaesthetized with chloral hydrate. The arteries were cut into segments of about 1.5cm. The endothelium of artery segments was injured with toothpick before being cultured. The artery segments were randomly divided into two experimental groups of cultured in containing CGRP(2×10-7mol/L )and no CGRP culture medium respectively,and one control group in which the artery segments had not been cultured in vitro. BrdU (8×10-4mol/L) was added into the culture medium before 24h of accomplishing samples. The proliferous cells were observed with HE staining and were labeled by BrdU with immunocytochemical method,and the mRNA expressions of HRG-1 and SM22αwas detected by RT-PCR.
