Objective: To purify the expression product of procaryon recombinant expression plasmid of human IL-24 in E coli, and observe the effects of recombinant human interleukin-24(rhIL-24) on the growth of gastric cancer cell line SGC-7901 in vitro. And to investigate the anti-tumor mechanisms of rhIL-24 on human gastric cancer cell line SGC-7901.Methods: IPTG was used to induce pET-21a(+)-IL-24 to express IL-24 in E.coli, after being purificated and renaturated, the rhIL-24 was obtained. Then rhIL-24 was used to react with human gastric cancer cell line SGC-7901 in vitro. The effects of rhIL-24 protein on gastric cancer cells in vitro was detected by MTT assay, apoptotic peak was measured by flow cytometry, and the morphous of apoptotic cells was observed by fluorescence staining. The expression of bax in human gastric cancer cell line SGC-7901 was detected by RT-PCR, Western Blotting, and the effect of rhIL-24 on chorioallantoic membrane was observed.Results: rhIL-24 was obtained and its purity coefficient is high. rhIL-24 can significantly inhibit the proliferation of gastric cancer cells in vitro which was detected by MTT assay, and its effects were dose-dependent and time-dependent. When the results were compared with the negative control group, the difference was significance(P<0.05). rhIL-24(30μg/ml) was used to react with SGC-7901, after 24h later the apoptotic peak was appeared by flow cytometry(FCM) and chromoplasm pyknosis was observed by fluorescence staining, we found the cell nucleus broke to pieces.
