Objective To identify gene expression profiles of gastric cancer and the suspect samples by endoscopic mucosal resection and to explore the molecular mechanism, clinical classification and subsequently the individual therapy of gastric cancer.Methods Tissue samples of highly suspect gastric cancer mucosa were obtained by endoscopic mucosal resection for biopsy. A total pairs of confirmed 28 gastric cancer and 19 precancerous tissue samples and their adjacent mucosa were collected and analyzed using spotted cDNA microarray containing 14592 gene elements. Filtering criteria was based on the presence of significant signal in≥80% of hybridizations. Hierarchical clustering was performed and viewed with TreeView. Discriminant genes and differences between groups were analyzed using Significant Analysis of Microarray. Validation of array results was carried out by real-time quantitative PCR (QPCR).Results 262 differentially regulated genes was identified in gastric cancer group, of which 30 regulated genes have the different characteristic expression pattern between the gastric cancer and non-gastric cancer samples. The result of hierarchical clustering show genes related to digestion and immune response have various expression within gastric cancer or non-gastric cancer groups. QPCR reaffirmed the array results.Conclusions Gene expression profiling of samples by endoscopic mucosal resection can provide a exact molecular pattern of transcriptome, which will benefit to probe into clinical diagnosis by molecular pathology, and some regulated genes has been identified to be candidate markers for gastric cancer, and distinct subtypes can be divided by different expressions of genes related to digestion and immune in both gastric cancer and non-gastric cancer groups.
