Objective The aim of this study was to observe the inhibition effects and the induction of differentiation on the human pancreatic cancer cells (PC-3) in vitro after induced by all-trans retinoic acid (ATRA) .At the same time, and expression change of Survivin was observed. Methods Experiment was divided into two groups: experimental group and control. In experimental group, PC-3 cells were cultured in nutrient fluid containing different density of ATRA, but the nutrient fluid contained no ATRA in control group. After induced 24, 48, 72, 96 hours, the inhibition rates of proliferation were detected by MTT assay, the change of Survivin mRNA expression level was examined by RT-PCR, flow cytometry(FCM) were used to detect the apoptotic rates and the cell cycle changes.Results (1) After induced by ATRA, The cell proliferation was significently inhibited in time-dependent and dose-dependent manners, the tendency of inhibition increased along with the dose and time gradual increased. (2)After induced by ATRA, the apoptotic rates of PC-3 cells were not obvious in the point of 24 hour, the apoptotic peak was observed from 48 hours, apoptotic rates achieved 11.01% in 96 hours.
