Objective: Gastric cancer is one of the most common malignacies in our country. Most of patients with this disease are diagnosed in advanced stages, and lost the chance of surgical treatment. Despite recent progress in chemotherapy, the overall survival rate of gastric cancer patients in advanced stages is still low. Apoptosis resistance of gastric cancer cell may be one of many reasons, and has increasingly become one of the hot spot of research fields. Inhibitor of apoptosis protein family may play a key role in cause apoptosis resisitance, and have become the focus of research increasingly. Livin is a novel inhibitor of apoptosis protein(IAP) family member, and it has two isoforms, Livin α and Livin β. The relationship between its up-regulation and gastric cancer occurrence, progress and Apoptosis remains uncertain. The aim of this study was to investigate the expression of Livin in gastric cancer, para-cancerous and benign disease gastric tissues, to explore relationship between Livin expression and clinical pathologic parameters, as well as the expression of Survivin in gastric cancer, and to investigate the apoptosis in gastric cancer SGC-7901 cell by siRNA-mediated silencing of the Livin gene.Methods: (1) The mRNA expression of Livin and Survivin were detected in malignant, para-cancerous, benign disease gastric tissues and gastric cancer cell lines respectively by reverse transcription polymerase chain reaction (RT-PCR).
