Tumorigenesis is not only related to the growing speed of cells, but also to the mortality speed of cells according recent researches. The defect or block of apoptosis is also an important factor that results in the occurrence and development of tumor. So cancer is the disease of abnormal proliferation and differentiation of cells, and the disease of abnormal apoptosis.Gastric cancer are one of the most common malignancies in Chinese population with its low early diagnosis rates ,The great challenge confronting us is its resistance to chemotherapy and radiotherapy in advanced stage. Many changes in molecular events including oncogenes ,growth factors, cyclins, tumor suppressor genes, cell adhesion molecules and unstability of genes are involved in the development and progression of gastric cancer. But cell proliferation and apoptosis constitutes the elementary process of tumorigenesis. As the new member of Inhibitor of Apoptosis Protein(IAP) gene family,Livin gene not only has characteristic of inhibiting apoptosis and promoting proliferation in tumor cells,But also increasing resistance to chemotherapy. It has great value to investigate in gene diagnose , treatment and drug fast.Objective:The purpose of study was to explore the expression of Livin gene in gastric cancer and para-cancerous and chronic gastritis tissues. With analysis of the relationships between the expression of Livin and degrees of differentiation, lymph node metastases and TNM stages. To investigate its possible mechanisms in the development and progression of gastric cancer . Aiming to find an ideal tumor marker for diagnosis and prognosis.Methods:1. 45 gastric cancer samples and 20 para-cancerous samples confirmed with histopathology were obtained from upper gastrointestinal surgery operation from 2006.6-2006.12. 30 chronic gastritis samples including ten specimens of chronic atrophic gastritis were also obtained from endoscopy. The mRNA expression levels of Livin were assessed by RT-PCR in these samples.2. 80 stomach carcinoma tissures and 30 chronic gastritis tissures including tenspecimens of chronic atrophic gastritis and 20 para-cancerous tissures the expression of Livin were examined by using Elivision method. All of the tissues were fixed in 10% neutral formalin and enbedded in paraffin collected from 2005.5 to 2006.6.The correlation between these expression and parameters features was analyzed.Result:1. Results from RT-PCR:1.1 In 45 gastric cancer tissues,the rate of positive expression of Livin gene was 48.8% (22/45). In contrast, tissues of superficial gastritis and atrophic gastritis and para-cancerous tissures did not express Livin. The discrepancy was significance statistically (P<0.01).1.2 Divide the expression rates using gastric cancer cell differentiation, The expression rate of Livin in moderately differentiated and well-differentiated carcinoma was25% by comparison to 68% in that of poorly differentiated and undifferentiatiated carcinoma. The discrepancy was significance statistically(P<0.05). The expression rate of Livin in Tl-T2was35.2% by comparison to 57.1% in that of T3-T4. The discrepancy was no significance statistically (P>0.0 5 ),But the positive rate of Livin expression was 34.7% in I+ II type(according to TNM grouping), and 63.6% in III + IV type,P>0.05. the positive rate of Livin expression was 60% in lymph node metastases,and 26.7% in no lymph node metastases, P<0.05,they has no significance statistically in age and gender(P>0.05).2. Results from Immunohistochemistry2.1 in contrast to no expression in chronic gastric mucosa and para-canceroustissures,Livin was expressed in 44 of 80(55%) cases of gastric cancer samples, The discrepancy was significance statistically(P<0.01).2.2 There was a relationship between Livin gene expression with degrees ofdifferentiation,lymph node metastases .they has no significance statistically in age and gender and infiltration and TNM stages.Conclusion:1. Compared to the no expression of Livin in para-cancerous tissures and chronic gastritis samples, The expression of Livin in gastric cancer tissues indicates that Livin gene facilitate the development in s gastric cancer.2. they has significance statistically in differentiated and lymph node metastases. Detection of Livin expression may provide a helpful judgement for prognosis.
