Objective: Neutron radiation can cause severe damages to the intestine which is hard to recover. Unfortunately, there is still no good cure so far. Therefore, in this study we explore the effect of IL-11 on neutron irradiated intestine epithelim injury and its signal transduction mechanism, which might help to find new potential therapies.Methods: 178 BALB/C mice were irradiated by 2.5 and 4.0 Gy neutron and injected subcutaneously with rhIL-11 at a dose of 600μg/kg body weight once per day for 3 days prior to or after irradiation. The mice were killed at 6h,1d, 2d, 3d, 5d, 7d, 14d and 28d after irradiation. IEC-6 cells were irradiated by 4.0 Gy neutron and treated with rhIL-11 12h prior to or immediately after irradiation. Effects of IL-11 on the intestinal histopathological changes, cellular biological behaviors, the expressions of IL-11, IL-11Rα and gpl30 and activities of JAK1 and STAT3 were observed by means of HE staining, IPCM, MTT, flow cytometry, immunohistochemistry, western blot, EMSA and image analysis.Results: (1) After 2.5 Gy neutron irradiation, the cryptal cell neucleus showed swelling, pyknosis and karuorrhexis in 2 days and regeneraton was observed at 3~7d after irradiation. Mice exposed to 4.0 Gy neutron irradiation showed more severe intestinal damages as compared with those exposed to 2.5 Gy neutron irradiation and couldn't cure spontaneously. IL-11-treated mice had a larger number of epithelial cell and higher villus. (2) Intense immunoreactivity of IL-11, IL-11Rα and gp130 was located within the cytoplasm of normal intestinal epithelium of the mice. At 6h after 2.5 Gy neutron irradiation, immunoreactivity of IL-11 and IL-11Rα increased, while at the first and second day after irradiation, their expressional levels decreased and came back to normal level at 7~14d after irradiation.
