Neutron-irradiation can cause more serious injury than r-ray does. rhIL-11 is a multi-effective cytokine and has been applied in stimulating hematopoietic recovery and protecting intestine from injury. In order to estimate the effects of rhIL-11 on neutron-induced ARS, we used bone marrow cell culture and FACS technology to observe the therapeutic effect of rhIL-11 on mouse hematopoietic system and intestine. We also investigated the effect of rhIL-11, when combined with G-CSF on hematopoietic recovery of mice and beagles irradiated by neutron 3.0Gy and 2.0Gy, respectively.The results revealed that rhIL-11 significantly improved proliferation of cells in mice injected subcutaneously either before or after irradiation with 3.5 Gy of 90% neutron, drove the bone marrow cells into cell cycle,shortened the proliferation period of intestinal cells. Administered before or after total body irradiation(TBI), rhIL-11 could remarkably enhance the survival rate of neutron-irradiated mice, improve the WBC counts within 24hr, increase the percentage of bone marrow CD34 positive cells and CFU-GM count. When rhIL-11 was combined with rhG-CSF the effect could be strengthened. The results from beagles showed that rhIL-11 itself was less effective on increasing WBC counts than rhG-CSF, alone or combined with rhIL-11. All of them had the effect of stimulating bone marrow cells to form more CFU-GM colonies. rhIL-11, when combined with rhG-CSF, could increase the percentage of bone marrow CD34 positive cells and the ratio of CD4+/CD8+ in peripheral blood. Histopathological observation suggested that combined application of rhIL-11 and rhG-CSF significantly accelerated the recovery of hematopoietic and immunological functions of neutron-irradiated beagles.
