Objective: To investigate the effect and mechanism of transforming growth factor β1 (TGF-β1) on the proliferation in SNU-601/cis2 multi-drug resistant gastric carcinoma cell line. Methods: The experiment was divided into two groups: control group andexperiment group. The control group referred to the SNU-601/cis2 cell line only, and no TGF-β1. The experiment group referred to the SNU-601/cis2 cell line added 10 ng/mL TGF-β1. All groups cell lines were respectively cultured for 24, 48, and 72h. The cell survival rate was detected by using MTT assay in all groups and time period, and the c-myc, mutant type p53 and cyclin D1 protein expressions were detected by using immunohistochemistry and Western Blotting techniques. Results: MTT assay showed that the growth inhibitory rate of SNU-601/cis2 multi drug resistant gastric carcinoma cells was increased along with the time and it had the concentration and time dependent. All were significantly different compared with the control group (P<0.001). Inverted microscope showed that the cell proliferation of control group was increased along with the time elongated, however, 10 ng/mL TGF-3 1 treated experiment group showed the cell proliferation was significantly decreased in all time period compared with the control group. The immunohistochemistry indicated that c-myc and mutant type p53 protein expressions were strongly positive in the control group cells at the time of 24, 48, and 72h, however, the expression of c-myc and mutant type p53...
