In clinical research the high sensitivity, wide dynamic range and of chemiluminescent assays has led to a vast range of applications, notably in immunoassays, protein and nucleic acid blotting, microarray- basedassays, capillary electrophoresis (CE), and flow-injectionanalysis. Meanwhile microfluidic devices have many advantages: miniaturization, high performance and low cost. So they have great potential in immunoassays environment protection and monitoring.In this research chemiluminescence immunoassay was carried out in biochip, In order to develop a fast, sensitive, low-cost assay. Especially, the study and application of this chemiluminescence immunoassay on the pathogeny , hormone and environmental analysis will be of great practice significance.Based on these points, the dissertation would lay stress on the study of new biochip magnetic chemiluminescence enzyme immunoassay methods. It consists of three chapters:Chapterl The principles and applications of chemiluminescence immunoassay are reviewed. Enzyme immunoassay (EIA) , chemiluminescence immunoassay (CLIA) and some new immunoassay were described here.And the development and applications of microfluidic chip devices are reviewed. More details are given on its materials, microfabricated technology and applications.Chapter2 A flow-type cell microchip was designed fabricated with glass. In the Chapter, a biochip magnetic chemiluminescence enzyme immunoassay for the determination of thyroid-stimulating hormone (TSH) based on the improvement of the method of the magnetic immunoenzyme spectrophotometric assay was established. The alkaline phosphatase (ALP) was used as label reagent, which reacted with the chemiluminescence reagent of 4-methoxy-4- (3-phosphatephenyl)-spiro-(1,2-dioxetane-3,2'-adamantane) (PPD) in buffer solution. The linear range was 0.094~50 mIU/L TSH; intraassay and inter-assay were both below 12.7%. The recoveries were in the range of 91~109%. Compared with other immunoassay, the correlation coefficient was 0.965 and0.98892. The sensitivity of the present chemiluminescent method was 0.094 mIU/L.Chapter3 Thyroxine(T4) was detected with this biochip magnetic chemiluminescence enzyme immunoassay. The linear range was 20~150 ng/l T4; intraassay and inter-assay were both below 5.86%. The correlation coefficient was 0.98859 while compared with other immunoassay, The sensitivity was 4.5 ng/l..
